01Weekend One

The petri dish. Form teams, ideate against the tracks, and pitch a one-week experiment.

  • Projects documented into the public gallery
Phase_01

The Prototype Weekend

Two days to turn a room of strangers into teams with a plan that's actually runnable at a BSL-1 bench in a week. Scope discipline here is the single biggest predictor of whether a team ships.

What happens

  • Friday: opening, tracks drop (e.g. Longevity × AI, Biosecurity, Wet-Lab Systems), team-formation mixer.
  • Saturday: ideation + feasibility workshops, mentor scoping, a "can you build this in a week?" gut-check with a lab lead.
  • Sunday: pitches to a panel; feedback; accepted teams matched to bench slots and reagent budgets.

Why the pitch is a gate

The pitch review is also the safety and feasibility gate. Judges and the lab's safety officer screen every proposal before it touches a bench — ruling out anything human-subject, clinical, animal, gain-of-function, or beyond BSL-1. A green-lit pitch is a promise the experiment is buildable, safe, and legal.

Phase_02

The Lab Week

The part nobody else does. Real bench access at a licensed BSL-1 lab — at Biopunk Lab HQ that's Frontier Tower's wet-lab floor, with $500k+ of gear — while sponsors cover the reagents that would otherwise make it impossible. A realistic one-cycle timeline:

Day 1 · Mon

Setup & prep

Pre-arrived synthetic DNA and the stock parts kit are waiting. Prep competent cells and media; finalize the build plan with a mentor.

Day 2–3 · Tue–Wed

Build

Assembly (Golden Gate / Gibson / restriction–ligation), transformation, overnight growth, colony picks and minipreps.

Day 4 · Thu

Validate

Send constructs for ~$15 overnight whole-plasmid sequencing. Set up the functional assay.

Day 5 · Fri

Test & read out

Confirm sequences; run the reporter / growth / biosensor assay; collect the data that becomes the demo.

Weekend

Analyze & tell the story

Plot results, build the demo, frame what you learned and what's next.

Buildable in a week

  • PCR, molecular cloning, bacterial/yeast transformation
  • Miniprep + overnight sequence validation
  • Small-scale reporter/protein expression & fluorescence assays
  • Simple biosensor prototypes; growth/inhibition assays
  • Basic CRISPR editing in bacteria or yeast

Ruled out at the gate

  • Mammalian stable cell lines; animal or mouse work
  • Human subjects — including self-experimentation
  • Directed evolution / multi-week passaging
  • Large-scale fermentation; structural biology
  • Anything above BSL-1, clinical, or select-agent
The deliverable The demo-day output is a proof-of-concept plus a plan — "the construct expresses / the strain grows / the sensor lights up" — not a finished product. iGEM's Design–Build–Test–Learn cycle, compressed into one honest sprint.
Phase_03

Demo Day

The second weekend. Teams present living data to the tribunal — founders, investigators, and operators from the local bio scene. Run it science-fair style: teams at numbered stations, judges circulating, every project seen by at least three.
